Counting cells with the hemocytometer

Get a printable version of this protocol (requires Adobe Acrobat

  1. Trypsinize the cells as described before.
  2. Place the coverslip on the hemocytometer slide.
  3. Allow one drop of cell suspension from a pasteur pipette to be drawn under the coverslip.
  4. Place on the microscope and count the 16 squares in each corner of the grid (see figure).
To calculate:

cells/ml =( total # cells)/4 x 104

The hemocytometer grid (circles indicate the 16 squares to be counted)
